Poke with a stick or thump it. This might sound daft but giving joints a prod with an insulated stick can reveal issues. Gently thumping an amp in various places does the same thing. so, try prodding grounds, valve pins etc. Another thing I do is to examine all my joints with a magnifying glass. I can't see detail even with glasses so a magnifying glass is a must.
I feel your pain here, and I'm completely lost as to where you are, I feel your just stumbling in the dark, your need to know more how your circuit is working, what wire does what etc, some reading at night's after you've finished would be very beneficial I think. If your getting really p*ssed off, walk away, put the amp away, forget the thing for a few weeks and then come back to it. no one likes giving up, but sometimes a tactical withdrawal gives you the space and distance to look at things afresh.
