If you go to the drive letter in Explorer, then right-click on it, it should offer you the option to eject or dismount the drive (can't remember the exact wording as I haven't used Vista EVER!).
The other thing to try is to press F5 whilst in the directory window - this should also refresh the cache. On Windows 7, right-clicking in the Explorer directory windows and selecting "Refresh" has the same effect...
